ABSTRACT

Abstract Present study was conducted to record ecotoparasitic prevalence in bat fauna of the northwestern parts of Pakistan. A total of 204 bat specimens representing 14 species were captured during a two year survey, extending from June 2015 through May 2016. A species of soft ticks Argas vespertilionis was identified from 23 bat specimens. Similarly, members of the family Dermanyssoidae (dermanyssoid mites) were isolated from 10 bat specimens, that of Spinturnicidae (spinturnicid mites) from 3 and Streblidae (bat flies) from 2 bat specimens. These parasites were collected using entomological tweezers and were identified on morphological basis. Further studies on parasitic prevalence, molecular characterization of bat parasites and their control measures are recommended.


Resumo O presente estudo foi realizado para registrar a prevalência de ectoparasitas na fauna de morcegos em partes do noroeste do Paquistão. Um total de 204 espécimes de morcegos, representando 14 espécies, foi capturado durante uma pesquisa de dois anos, de junho de 2015 a maio de 2016. A espécie de carrapato Argas vespertilionis foi identificada em 23 espécimes de morcegos. Da mesma forma, os membros da família Dermanyssidae (ácaros dermanyssoid) foram isolados de 10 espécimes de morcego, os da Spinturnicidae (ácaros spinturnicid), de 3, e os da Streblidae (mosca de morcego), de 2 espécimes de morcego. Esses parasitas foram coletados com pinça entomológica e identificados com base morfológica. Estudos adicionais sobre prevalência parasitária, caracterização molecular de parasitas de morcego e suas medidas de controle devem ser realizados.

Resumen Este informe describe el hallazgo de garrapatas blandas (Ixodida, Argasidae) parasitando a dos carpinteros andinos (Colaptes rupicola) provenientes del distrito de Nuñoa, provincia de Melgar en Puno, Perú (14°31'11.77"S; 70°32'15.95"W; 3967 m de altitud). Un total de 29 larvas de garrapatas fueron colectadas directamente de las aves y posteriormente identificadas morfológicamente como Argas neghmei. El diagnóstico fue basado en la morfología de las larvas y confirmado mediante el análisis molecular del gen 16S ARNr mitocondrial de la garrapata. Un fragmento de 363 pares de bases de este gen mostró una identidad del 100% con secuencias registradas para A. neghmei de Argentina y Chile (GenBank: FJ853598 y DQ295781). Este hallazgo confirma la presencia de A. neghmei en Perú y agrega al carpintero andino como nuevo hospedero para esta garrapata.


Abstract This report describes the finding of soft ticks (Ixodida, Argasidae) parasitizing two Andean flickers (Colaptes rupicola) from the Nuñoa district, Melgar province in Puno, Peru (14°31'11.77"S; 70°32'15.95''W; elevation 3967 m). A total of 29 larval ticks were collected directly from the birds and morphologically identified as Argas neghmei. Diagnosis was based on the larval morphology and confirmed by molecular analysis of the tick mitochondrial 16S rRNA gene. A sequence of 363 base pair of this gene showed to be 100% identical with sequences of A. neghmei from Argentina and Chile (GenBank: FJ853598 and DQ295781). This finding confirms the presence of A. neghmei in Peru and adds the Andean flicker as a new host for this species.

ABSTRACT

The pigeon tick Argas reflexus is a pathogen-transmitting soft tick that typically feeds on pigeons, but can also attack humans causing local and systemic reactions. Chemical control is made difficult due to environmental contamination and resistance development. As a result, there is much interest in increasing the role of other strategies like biological control. In this study, the efficacy of three strains (V245, 685 and 715C) of entomopathogenic fungus Metarhizium anisopliae for biological control of three life stages of pigeon tick A. reflexus including eggs, larvae, engorged and unfed adults was investigated under laboratory conditions. Five concentrations of different strains of M. anisopliae ranging from 10³ to 10(7) conidia/ml were used. All fungal strains significantly decreased hatchability of A. reflexus eggs. Strain V245 was the most effective strain on the mortality of larval stage with nearly 100 percent mortality at the lowest concentration (10³ conidia/ml) at 10 days post-inoculation. The mortality rate of both engorged and unfed adult ticks were also increased significantly exposed to different conidial concentrations compared to the control groups (P < 0.05) making this fungus a potential biological control agent of pigeon tick reducing the use of chemical acaricides.

ABSTRACT

Os parâmetros do ciclo biológico de Argas (Persicargas) miniatus, fase adulta, foram estudados em condições controladas (27±1°C e 80±10 por cento UR) e em condições ambientais não controladas (ambiente de laboratório nas estações chuvosa e seca). Nessas condições, os machos se originaram preferencialmente de N2 e as fêmeas de N3. As fêmeas realizaram até 18 posturas (em condições controladas), enquanto que as fêmeas mantidas nas estações chuvosa e seca realizaram até 9 e 12 posturas, respectivamente. O número total de ovos foi 1350 (em condições controladas), 443 ovos na estação chuvosa e 894 ovos na estação seca. Em jejum, os carrapatos machos sobreviveram por 165 dias (condições controladas) e 135 dias em ambiente de laboratório; as fêmeas sobreviveram por 300 dias em condições controladas e 240 dias no ambiente de laboratório. Nas condições ambientais não controladas, os parâmetros biológicos do estágio adulto de Argas (Persicargas) miniatus sofreram alterações negativas, principalmente durante a estação chuvosa.


The life cycle of Argas (Persicargas) miniatus in adult stage was studied in controlled conditions (27±1°C and 80±10 percent RH) and uncontrolled temperature and humidity conditions (room conditions during wet and dry season). Males originated preferentially from N2 and females from N3. Females in controlled conditions laid eggs up to 18 times, while females in uncontrolled room conditions, during wet and dry seasons, laid eggs 9 to 12 times, respectively. In controlled conditions, females laid 1350 eggs, whereas in uncontrolled conditions females laid 443 eggs during the wet season and 894 eggs during the dry season. Male ticks survived without feeding for 165 days in controlled conditions and 135 days in uncontrolled conditions, while females survived for 300 days in controlled conditions and 240 days in uncontrolled conditions. In ambient conditions, biological parameters of Argas (Persicargas) miniatus adult stage were negatively affected especially during the wet season when compared with adult stage kept in controlled condition.

ABSTRACT

Background & objectives: Crimean-Congo hemorrhagic fever (CCHF) virus is a tick-borne member of the genus Nairovirus, family Bunyaviridae. CCHFV has been isolated from at least 31 different tick species. The virus is transmitted through the bite of an infected tick, or by direct contact with CCHFV-infected patients or the products of infected livestock. This study was undertaken to study the genetic relationship and distribution of CCHFV in the tick population of Hamadan province of Iran. Method: In this study, RT-PCR has been used for detection of the CCHFV genome. Results: This genome was detected in 19.2% of the ticks collected from livestock of different regions of the Hamadan province in western Iran. The infected species belonged to Hyalomma detritum, H. anatolicum, Rhipicephalus sanguineus and Argas reflexus. With one exception, genetic analysis of the virus genome isolates showed high sequence identity to each other. Even though they clustered in the same group with the strain circulating in Iran, they had a closer relationship to the Matin strain. Interpretation & conclusion: Vector control programs should be applied for reducing population density of potential tick vectors in this province. Further surveys are indicated in this region to provide a better view of the distribution and epidemiology of the virus.

ABSTRACT

The current study investigated the biology of nymphs of the first and second instars of Argas (Persicargas) miniatus. Nymphs were deprived of food for 15, 30 or 60 days and held at 27 ± 1 ºC and 80 ± 10 percent relative humidity (controlled conditions) or at room conditions of temperature and relative humidity. Nymphs of first instar deprived of food for 15 or 30 days molted to second and third instars in both controlled and room conditions. Nymphs of the first instar deprived of food for 60 days had 28 and 37 percent mortality in controlled and room conditions, respectively; and survivors did not attach to the host. Nymphs of the second instar, deprived of food for 60 days, molted either to the third instar or to males after feeding on Gallus gallus, and the nymphs of the third instar developed to adults (42.42 percent males and 36.36 percent females when nymphs were held in controlled temperature and humidity conditions, and 40.54 percent males and 48.65 percent females when nymphs were held in room conditions). The remainder of the nymphs molted to the fourth instar and then molted to females. In conclusion, the nymphal starvation period of 60 days determined the number of nymph instars in the life cycle of A. miniatus under the experimental conditions studied.


Os aspectos biológicos de ninfas de primeiro e segundo instares de Argas (Persicargas) miniatus quando submetidas a diferentes períodos de jejum (15, 30 e 60 dias), foram estudados em estufa climatizada (27 ± 1 ºC e 80 ± 10 por cento de umidade relativa) e em ambiente de laboratório. Ninfas de primeiro instar que foram submetidas a um período de jejum de 15 e 30 dias mudaram para ninfas de segundo e terceiro instar, em ambas as condições estudadas. No período de 60 dias de jejum verificou-se mortalidade de 28 e 37 por cento das ninfas de primeiro instar, em estufa climatizada e em ambiente de laboratório, respectivamente. As ninfas sobreviventes não se fixaram sobre os hospedeiros. As ninfas de segundo instar, após 60 dias de jejum, desenvolveram-se em ninfas de terceiro instar ou machos, quando alimentadas em Gallus gallus. Ainda neste grupo, as ninfas de terceiro instar mudaram para adultos (42,42 e 40,54 por cento machos; 36,36 e 48,65 por cento fêmeas, nas condições ambiente de laboratório e estufa climatizada, respectivamente) e o restante desenvolveu-se em ninfas de quarto instar que por sua vez mudaram para fêmeas. Então, a situação de jejum (60 dias) em que as ninfas foram submetidas determinou o número de ninfas no ciclo biológico de A. miniatus, sob as condições experimentais estudadas.

ABSTRACT

In order to describe the developmental biology of the tick Argas neghmei Kohls & Hoogstraal under laboratory conditions, 40 females and 40 males were collected from chicken coops located in Calama (II Region, Chile). They were fed on chickens and maintained under two laboratory conditions: one group at 30 ± 5 ºC and 35 ± 5 percent RH and another at 27 ± 5 ºC and 80 ± 5 percent RH, both at 12: 12 h L:D photoperiod. The ticks were observed daily to determine larval feeding periods, preoviposition, oviposition, egg incubation as well as the frequency of egg laying, number of eggs laid, and percentage of larval hatching. Females did not lay eggs at 80 ± 5 percent RH, and data on the biology of this tick was obtained only at 35 ± 5 percent RH. The life cycle of A. neghmei lasted an average of 269 days. Feeding period of each nymphal stage as well as of adult females between oviposition events lasted less than a day. Females laid on average 1.8 egg batches and egg-laying period lasted on average 14 days, during which about 96 eggs were laid per female.

ABSTRACT

Borreliose aviária é uma doença septicêmica aguda, cosmopolita, que acomete diferentes espécies aviárias, sendo causada por Borrelia anserina Sakharoff, 1891. O objetivo do presente trabalho foi avaliar as alterações hematológicas em Gallus gallus domesticus experimentalmente infectados por B. anserina via vetor Argas (Persicargas) miniatus. Um total de 27 aves da espécie G. g. domesticus com 67 dias de vida, foram divididas em três grupos inteiramente casualizados contendo nove animais cada. Um grupo foi exposto a carrapatos infectados por B. anserina (Grupo 1); outro a carrapatos livres deste agente (Grupo 2); além de um grupo não exposto aos carrapatos (Grupo 3). Realizaram-se esfregaços sangüíneos diariamente, a partir do primeiro dia de exposição ao vetor, até o 25º dia pós-exposição (DPE). Amostras de sangue foram coletadas 3 dias antes da exposição aos carrapatos, e 3, 8 e 18 dias pós-exposição (DPE), para a realização dos hemogramas. O exame dos esfregaços das aves do Grupo 1 revelou grande número de espiroquetas. Os esfregaços sangüíneos dos Grupos 2 e 3 mantiveram-se negativos durante todo o período experimental. De acordo com os resultados das avaliações hematológicas, as aves do Grupo 1 apresentaram um quadro de anemia normocítica normocrômica em oito DPE, além de leucocitose com heterofilia e monocitose que cursaram paralelamente ao período de espiroquetemia. Concluiu-se que a infecção por B. anserina determinou nas aves do Grupo 1 alterações hematológicas compatíveis com uma infecção bacteriana de moderada gravidade, evoluindo para auto-cura, nas condições experimentais estabelecidas neste trabalho.(AU)


Avian spirochaetosis is a cosmopolite acute septicemic disease of many avian species, caused by Borrelia anserina Sakharoff, 1891. The present study assesses the estimate of the hematological alterations of Gallus gallus domesticus experimentally infected with B. anserina by vector Argas (Persicargas) miniatus. Twenty-seven fowls of the species G. g. domesticus, 67 days old, were randomly allocated into three groups composed by nine animals each. One group was exposed to B. anserina infected ticks (Group 1), other one to ticks free of this agent (Group 2), and another group not exposed to ticks (Group 3). Blood smears of the fowls were taken daily, since the first day the fowls were exposed to the ticks, up to the 25th day after exposure (DAE). Blood samples were collected three days before exposure, and three, eight and 18 DAE, for hematologic tests. The examination of Group 1 smears revealed large number of spirochaetes. Group 2 and 3 blood smears were negative during the whole period under exam. In agreement with the hematological evaluation results, the fowls exposed to infected ticks showed a normocytic normochromic anemia in eight DAE, leucocytosis with heterophilia and monocytosis concomitant with the spirochaetemia. We concluded that B. anserina infection determined on fowls of Group 1 hematological alterations compatible with bacterial infection of moderate gravity, developing to self-cure, in the experimental conditions established in this study.(AU)

Foram coletados 665 espécimes de Argas miniatus em dois municípios dos estados da Bahia e Minas Gerais. Destes, 596 (89,6 por cento) tinham se alimentado, sendo que 489 (82 por cento) reagiram para um único tipo de sangue, distribuídos entre aves (46,8 por cento), roedores (30,9 por cento), gambás (14,5 por cento), bovinos (4,3 por cento) e eqüinos (3,5 por cento). Nesse tipo de reação, o sangue de mamíferos foi detectado em 53,2 por cento (260/489) dos argasídios. As reações múltiplas foram observadas em 107 (17,9 por cento) carrapatos, com sangue de aves presente em 84,1 por cento (90/107), enquanto, o sangue de mamíferos reagiu em 100 por cento (107/107). Os resultados apontam para a inespecificidade parasitária


Six hundred and sixty-five specimens of Argas miniatus were collected in two municipalities of Bahia and Minas Gerais states, Brazil. Five hundred and ninety-six (89.6 percent) of them had fed and 489 (82 percent) of them reacted to only one type of blood, including birds (46.8 percent), rodents (30.9 percent), opossuns (14.5 percent), bovines (4.3 percent) and horses (3.5 percent). In that reaction, the type of mammal blood was detected in 53.2 percent (260/489) of the ticks. Multiple reactions were observed in 107 (17.9 percent) ticks, with blood of birds present in 84.1 percent (90/107), while the blood of mammals was detected in 100 percent (107/107). The results point for the nonspecificity of parasitism

Nympha from the first instar to the fifth consist of 4, 5, 5, 4 and 4 bands respectively.Esterase isoenzyme of the male and female consists of C and 6 bands respectivly. Nympha from the first instar to the fifth consist of 3, 6, 4, 6 and 7 bands respectively.Lactic dehydrogensse isoenzyme of the male and female consists 2 and 2 bands respectively.
